Applications | |
---|---|
Application | This Anti-JAK1 Antibody is validated for use in IP, WB for the detection of JAK1. |
Key Applications |
|
Application Notes | Immunoprecipitation: Use at 5-10μl per 1-2mg lysate Western blot: (1:1000-1:2000) The amount of JAK1 in cell lines tends to be extremely low. If the signal is weak it is recommended that a RIPA cell lysate (1-2mg protein) be prepared and JAK1 be first concentrated by immunoprecipitation and then immunoblotted. A degradation product of JAK1 (~110kDa) is also detected by the antiserum in immunoblots. Optimal working dilutions must be determined by end user. |
Biological Information | |
---|---|
Immunogen | Synthetic peptide corresponding to 21 amino acids of human/mouse/rat JAK1 (KTLIEKERFYESRCRPVTPS-C) |
Host | Rabbit |
Specificity | Recognizes JAK1 (Janus Kinase 1). The members of the JAK family of cytoplasmic protein tyrosine kinases physically associate with ligand-bound receptors. It is this association that results in tyrosine phosphorylation and activation. JAK1 is approximately 130 kDa and contains a C-terminal tyrosine kinase domain, an adjacent kinase or kinase-related domain, and five other domains that are highly conserved among JAK family members. |

Entrez Gene Number |

Entrez Gene Summary | Janus kinase 1 (JAK1), is a member of a new class of protein-tyrosine kinases (PTK) characterized by the presence of a second phosphotransferase-related domain immediately N-terminal to the PTK domain. The second phosphotransferase domain bears all the hallmarks of a protein kinase, although its structure differs significantly from that of the PTK and threonine/serine kinase family members. JAK1 is a large, widely expressed membrane-associated phosphoprotein. JAK1 is involved in the interferon-alpha/beta and -gamma signal transduction pathways. The reciprocal interdependence between JAK1 and TYK2 activities in the interferon-alpha pathway, and between JAK1 and JAK2 in the interferon-gamma pathway, may reflect a requirement for these kinases in the correct assembly of interferon receptor complexes. These kinases couple cytokine ligand binding to tyrosine phosphorylation of various known signaling proteins and of a unique family of transcription factors termed the signal transducers and activators of transcription, or STATs. |

UniProt Summary | FUNCTION: SwissProt: P23458 # Tyrosine kinase of the non-receptor type, involved in the IFN-alpha/beta/gamma signal pathway. Kinase partner for the interleukin (IL)-2 receptor. SIZE: 1142 amino acids; 131957 Da SUBUNIT: Interacts with IL31RA and SHB. SUBCELLULAR LOCATION: Intracytoplasmic membrane; Peripheral membrane protein. Note=Wholly intracellular, possibly membrane associated. DOMAIN: SwissProt: P23458 Possesses two phosphotransferase domains. The second one probably contains the catalytic domain (By similarity), while the presence of slight differences suggest a different role for domain 1. SIMILARITY: Belongs to the protein kinase superfamily. Tyr protein kinase family. JAK subfamily. & Contains 1 FERM domain. & Contains 1 protein kinase domain. & Contains 1 SH2 domain. |
