Background Information | CD117 is a 145 kDa transmembrane protein which belongs to the class III receptor tyrosine kinase family. The extracellular region consists of five immunoglobulin-like domains where the second and third loops are thought to be involved in ligand binding (Ashman, 1997; Bühring, 1995). CD117 has been shown to be a marker for tissue mast cells, hematopoietic stem cells, and progenitor cells in normal human bone marrow. The majority of CD117-positive marrow cells (50-70%) co-express CD34 and comprise progenitor cells and their precursors of all hematopoietic lineages. Apart from mast cells, CD117 is essentially absent from mature hematopoietic cells. Although CD117 is primarily a marker for non-lymphoid progenitor cells in the hematopoietic cells/tissues, including melanocytes, embryonic brain, and the reproductive system (Ashman, 1997). Anti-CD117, 104D2, was included in the Sixth International Workshop and Conference on Human Leucocyte differentiation Antigens, and studies by different laboratories confirmed its reactivity with CD117 (Ashman, 1997). |
